Maiorka,A; Silva,AVF; Santin,E; Dahlke,F; Bruno,LDG; Boleli,IC; Macari,M; Trautenmuller,H.
ABSTRACT The aim of this study was to evaluate the possible effects of glutamine and broiler breeder age on the development of intestinal mucosa in broiler chicks during first week of age. For this purpose, 32 one-day-old broiler chicks were distributed according to a completely randomized experimental design in a 2 x 2 factorial arrangement. Treatments consisted of two broiler breeder ages (30 and 60 weeks) and two dietary glutamine supplementation levels (0% and 1%). The morphological development of intestinal mucosa, particularly villus height and crypt depth, was evaluated. The results showed that the supplementation with 1% of glutamine influenced the development of villus height in the duodenum (p=0.009), jejunum (p = 0.006), and ileum (p = 0.001),...

Wu,Q; Liu,Z; Li,S; Jiao,C; Wang,Y; Wang,Y.
ABSTRACT The aim was to investigate the effect of glutamine (Gln) on broilers challenged with Salmonella Enteritidis. 240 1-day-old birds were divided into four groups in a completely randomized design, each of which included 6 replicates with 10 birds per replicate. Group I served as the unchallenged, untreated control (CON). All birds in groups II (SCC) - IV were challenged with 2.0 × 104 CFU/mL of S. Enteritidis. Birds in group III and IV were treated with 0.5% (Gln 1) and 1.0% (Gln 2), respectively, of Gln. The results indicated that S. Enteritidis infection led to a decrease in the average body weight at d 7, 14, and 21 (p<0.05). Chickens fed the Gln showed improved average body weights in comparison with the SCC group (p<0.05). At d 4, 7, 14,...

Glutamine and glutamate as vital metabolites BJMBR
Newsholme,P.; Lima,M.M.R.; Procopio,J.; Pithon-Curi,T.C.; Doi,S.Q.; Bazotte,R.B.; Curi,R..
Glucose is widely accepted as the primary nutrient for the maintenance and promotion of cell function. This metabolite leads to production of ATP, NADPH and precursors for the synthesis of macromolecules such as nucleic acids and phospholipids. We propose that, in addition to glucose, the 5-carbon amino acids glutamine and glutamate should be considered to be equally important for maintenance and promotion of cell function. The functions of glutamine/glutamate are many, i.e., they are substrates for protein synthesis, anabolic precursors for muscle growth, they regulate acid-base balance in the kidney, they are substrates for ureagenesis in the liver and for hepatic and renal gluconeogenesis, they act as an oxidative fuel for the intestine and cells of the...

Influence of genotype on protein and oil concentration of soybean seeds BABT
Santos,Esmael Lopes dos; Pípolo,Antonio Eduardo; Faria,Ricardo Tadeu de; Prete,Cássio Egidio Cavenaghi.
Aiming at evaluating genotype influence on the concentration of protein and oil, immature seeds of cultivars CD 202 and CD 206 were removed from the mother-plant, in the stage R5, and were grown in vitro, in a liquid culture medium which contained 20, 40 and 60 mM of glutamine, during eight days. Afterwards, the concentrations of oil and protein were compared to the contents of the seeds cultivated in vivo. With a higher availability of glutamine for the seed, there was an increase of protein content. The genotypes were statistically different as far as the protein concentration was concerned, which confirmed that the genotype had influence on the concentration of protein in the seed. Oil and protein concentrations were inversely related when a variation of...

Metabolic fate of glutamine in lymphocytes, macrophages and neutrophils BJMBR
Curi,R.; Newsholme,P.; Pithon-Curi,T.C.; Pires-de-Melo,M.; Garcia,C.; Homem-de-Bittencourt Jr.,P.I.; Guimarães,A.R.P..
Eric Newsholme's laboratory was the first to show glutamine utilization by lymphocytes and macrophages. Recently, we have found that neutrophils also utilize glutamine. This amino acid has been shown to play a role in lymphocyte proliferation, cytokine production by lymphocytes and macrophages and phagocytosis and superoxide production by macrophages and neutrophils. Knowledge of the metabolic fate of glutamine in these cells is important for the understanding of the role and function of this amino acid in the maintenance of the proliferative, phagocytic and secretory capacities of these cells. Glutamine and glucose are poorly oxidized by these cells and might produce important precursors for DNA, RNA, protein and lipid synthesis. The high rate of...

Modulation of murine blastocyst hatching in vitro by glutamine and tryptophan BJMBR
Castro,W.; Bueno Sánchez,J.C.; Piedrahita-Ochoa,C.; Cadavid J.,A.P..
Enrichment of culture media with amino acids improves embryo development. However, little is known about the specific action of each amino acid during embryogenesis. The present study was undertaken to examine the effect of L-glutamine (Gln) and tryptophan (Trp) on mouse embryo hatching, expansion and viability in vitro. Blastocysts were collected from 6- to 8-week-old female BALB/c mice (N = 30) and cultured in M2 medium containing either 0.125, 0.25 or 0.5 mM Trp, 1 mM Gln, or M2 alone. Gln significantly increased (100%; P < 0.05) blastocyst hatching at 24 h compared to M2 alone or Trp; moreover, Trp inhibited blastocyst hatching when compared to M2 alone (P < 0.05) at 72 h. In contrast, the percentage of embryos reaching the state of expanded...

Ornithine decarboxylase expression in the small intestine of broilers submitted to feed restriction and glutamine supplementation Rev. Bras. Ciênc. Avic.
Fischer da Silva,AV; Borges,SA; Maiorka,A; Givisiez,PEN; Rocha,C; Macari,M.
Six hundred and forty one-day-old Cobb male broilers were used to evaluate ornithine decarboxylase (ODC) expression in the mucosa of the small intestine. Birds were submitted to early feed restriction from 7 to 14 days of age. The provided feed was supplemented with glutamine. A completely randomized design with a 2 x 2 factorial arrangement was used (with or without glutamine, with or without feed restriction). Restricted-fed birds were fed at 30% the amount of the ad libitum fed group from 7 to 14 days of age. Glutamine was added at the level of 1% in the diet supplied from 1 to 28 days of age. Protein concentration in the small intestine mucosa was determined, and ODC expression at 7, 14, 21, and 28 days of age was evaluated by dot blotting. ODC was...

Rat liver responsiveness to gluconeogenic substrates during insulin-induced hypoglycemia BJMBR
de Souza,H.M.; Borba-Murad,G.R.; Ceddia,R.B.; Curi,R.; Vardanega-Peicher,M.; Bazotte,R.B..
Hepatic responsiveness to gluconeogenic substrates during insulin-induced hypoglycemia was investigated. For this purpose, livers were perfused with a saturating concentration of 2 mM glycerol, 5 mM L-alanine or 5 mM L-glutamine as gluconeogenic substrates. All experiments were performed 1 h after an ip injection of saline (CN group) or 1 IU/kg of insulin (IN group). The IN group showed higher (P<0.05) hepatic glucose production from glycerol, L-alanine and L-glutamine and higher (P<0.05) production of L-lactate, pyruvate and urea from L-alanine and L-glutamine. In addition, ip injection of 100 mg/kg glycerol, L-alanine and L-glutamine promoted glucose recovery. The results indicate that the hepatic capacity to produce glucose from gluconeogenic...

Sources of trophic action on performance and intestinal morphometry of broiler chickens vaccinated against coccidiosis Rev. Bras. Ciênc. Avic.
Sakamoto,MI; Faria,DE; Nakagi,VS; Murakami,AE.
The aim of this experiment was to evaluate the effect of sources of trophic action (glutamine alone, glutamine associated with glutamic acid and yeast) associated with vaccination against coccidiosis on the performance and morphometry of the small intestine of broilers. In the trial, 1,200 broiler chicks were distributed according to a completely randomized design with a 3 x 2 + 2 (trophic action x vaccination or not + control - free trophic factor) factorial arrangement, with five replicates of 30 birds each. Vaccination negatively affected performance parameters and the morphometry of the intestinal mucosa, but at the end of the experimental period, among the broilers vaccinated against coccidiosis, the group fed glutamine presented better recovery from...

Understanding the mechanisms of glutamine action in critically ill patients Anais da ABC (AABC)
Oliveira,Gisele P.; Dias,Cristina M.; Pelosi,Paolo; Rocco,Patricia R.M..
Glutamine (Gln) is an important energy source and has been used as a supplementary energy substrate. Furthermore, Gln is an essential component for numerous metabolic functions, including acid-base homeostasis, gluconeogenesis, nitrogen transport and synthesis of proteins and nucleic acids. Therefore, glutamine plays a significant role in cell homeostasis and organ metabolism. This article aims to review the mechanisms of glutamine action during severe illnesses. In critically ill patients, the increase in mortality was associated with a decreased plasma Gln concentration. During catabolic stress, Gln consumption rate exceeds the supply, and both plasma and skeletal muscle pools of free Gln are severely reduced. The dose and route of Gln administration...

Variation in phytate accumulation in common bean (Phaseolus vulgaris L.) fruit explants BABT
Coelho,Cileide Maria Medeiros; Bellato,Cláudia Mattos; Garcia,Andréia Karime Marcelino; Vitorello,Victor Alexandre; Azevedo,Ricardo Antunes.
The in vitro synthesis of phytate was studied in common bean fruit explants. Different concentrations of sucrose; phosphorus (P); myo-inositol; abscisic acid (ABA); glutamine and methionine, were tested. Fixed concentrations of these compounds were tested at different periods (0, 3, 6 and 9 days). Variation in phytate coincided with different concentrations of sucrose, myo-inositol, P and ABA for the duration tested. These compounds caused an accumulation of phytate and were more effective in the presence of myo-inositol and P. The accumulation of P varied less than phytate for the different treatments tested in vitro. In conclusion, P, sucrose, ABA, and myo-inositol caused an increase in the phytate of bean seed, showing that it could be possible to alter...
